Chlorine is a critical anion involved in maintaining cellular homeostasis across various biochemical and physiological processes. Alterations in intracellular chloride (Cl−) concentrations influence key processes such as replication, transcription, and translation, which in turn affect cell cycle regulation, cellular proliferation, protein synthesis, and the maintenance of cellular volume and pH gradients. Intracellular Cl− also modulates metabolic activities within organelles. In the extracellular environment, Cl− is essential for preserving electrolyte balance within the blood, plasma, and interstitial fluid compartments. Recent advances in the understanding of Cl−-mediated cellular signaling have provided valuable insights into the molecular and metabolic changes associated with pathophysiological conditions characterized by dysregulated Cl− levels. This chapter will examine the most recent findings regarding the physiological and biochemical roles of Cl−, highlighting its significant contribution to human nutrition.
