Özet

Bu çalışma, et ve et ürünlerinde kalite kontrolü, tüketici sağlığı, dini inançlar ve ekonomik etik gereksinimleri doğrultusunda et türü tespitinin önemini ve bu amaçla kullanılan analitik metotları kapsamlı bir şekilde incelemektedir. Artan gıda fiyatları ve gıda ticaretinin küreselleşmesi ile birlikte işlenmiş et ürünlerinde ucuz et ikamesi, sakatat veya bitkisel protein katılması ve hatalı etiketleme gibi taklit ile tağşiş vakaları dünya genelinde yaygınlaşmıştır. Geleneksel fiziksel, anatomik, histolojik, kimyasal, serolojik/biyolojik (ELISA, presipitasyon) ve elektroforetik (PAGE, SDS-PAGE, IEF) ile spektroskopik (FT-IR, NIRS, NMR) yöntemler etiket doğrulamasında kullanılmakla birlikte, yüksek sıcaklık, pişirme ve işleme şartlarında proteinlerin denatüre olması sebebiyle işlenmiş veya karışık et ürünlerinde yetersiz kalabilmektedir. Bu teknik kısıtlamaları aşmak adına geliştirilen DNA tabanlı moleküler yöntemler; yüksek ısı, basınç ve ağır kimyasal işlemlere karşı son derece dirençli moleküler yapıları sayesinde üstün duyarlılık, doğruluk ve özgüllük sağlamaktadır. Polimeraz Zincir Reaksiyonu (PCR), türe özgü PCR, Multipleks PCR, Gerçek Zamanlı PCR (RT-PCR), Sanger ve Yeni Nesil Dizileme (NGS) ile izotermal amplifikasyon (LAMP, CPA, RPA) teknikleri; sığır, koyun, keçi, at, eşek, domuz ve kanatlı etlerinin yanı sıra cinsiyet ve tür orijininin hassas şekilde belirlenmesine imkan tanımaktadır. Sonuç olarak moleküler analizler, gıda denetim otoriteleri ile yetkili laboratuvarlar tarafından et ürünlerinde tağşişin önlenmesi, tüketici haklarının korunması ve yasal mevzuatlara tam uyumun sağlanmasında günümüzün en güvenilir yaklaşımı olarak öne çıkmaktadır.

This study comprehensively examines the importance of meat species identification in meat and meat products in line with quality control, consumer health, religious beliefs, and economic-ethical requirements, as well as the analytical methods used for this purpose. Driven by rising food prices and the globalization of food trade, instances of adulteration and mislabeling—such as substituting high-value meats with cheaper alternatives, offal, or plant proteins—have become widespread globally. Although traditional physical, anatomical, histological, chemical, serological/biological (ELISA, precipitation), electrophoretic (PAGE, SDS-PAGE, IEF), and spectroscopic (FT-IR, NIRS, NMR) methods are employed for label verification, they often prove insufficient in processed or mixed meat products due to protein denaturation under high temperature, cooking, and processing conditions. To overcome these technical limitations, DNA-based molecular methods provide superior sensitivity, accuracy, and specificity thanks to their highly resistant molecular structures against high heat, pressure, and harsh chemical treatments. Polymerase Chain Reaction (PCR), species-specific PCR, Multiplex PCR, Real-Time PCR (RT-PCR), Sanger and Next-Generation Sequencing (NGS), and isothermal amplification (LAMP, CPA, RPA) techniques enable the precise determination of gender and species origin as well as the detection of cattle, sheep, goat, horse, donkey, pork, and poultry meats. Consequently, molecular analyses emerge as today's most reliable approach utilized by food control authorities and authorized laboratories to prevent adulteration in meat products, protect consumer rights, and ensure full compliance with legal regulations.
