Bağlantı ve Rekombinasyon
Özet
Mendel'in bağımsız dağılım yasası, yalnızca farklı kromozomlarda ya da aynı kromozom üzerinde birbirinden yeterince uzakta yer alan genler için geçerlidir. Bu bölüm, aynı kromozom üzerindeki genlerin birlikte kalıtılması olgusunu (bağlantı) ve bu birlikteliğin krosing-over yoluyla kısmen bozulmasını (rekombinasyon) kapsamlı biçimde ele almaktadır. Bateson, Saunders ve Punnett'in Lathyrus odoratus çaprazlaması ki-kare testiyle çözümlenerek bağlantının ilk kanıtları gösterilmekte; Morgan'ın Drosophila melanogaster deneyleri, bağlantı grupları ve bulguların kromozom teorisiyle ilişkisi tartışılmaktadır. Tam ve eksik bağlantı, cis/trans konfigürasyonu, test çaprazlaması ve rekombinasyon frekansının harita birimine (centiMorgan) dönüştürülmesi sayısal örneklerle açıklanmaktadır. Krosing-overin mayoz sırasındaki sitolojik temeli (kiyazma, tetrat), iki kromatid ve dört kromatid modellerinin karşılaştırılması ve Neurospora tetrat analizleriyle doğrulanması ele alınmakta; çoklu ve çift krosing-overlerin harita mesafesini olduğundan küçük göstermesi ile Haldane haritalama fonksiyonu tartışılmaktadır. Üç noktalı çaprazlama üzerinden gen sırasının belirlenmesi, bölge bazlı rekombinasyon frekansları, koinsidans katsayısı ve interferens hesabı adım adım işlenmektedir. Bölüm, genetik ve fiziksel haritalar arasındaki farklar (sıcak-soğuk bölgeler, cinsiyet etkisi), moleküler işaretleyiciler, bağlantı dengesizliği haritalaması, GWAS ve genomik seleksiyon ile tamamlanmaktadır.
Mendel's law of independent assortment holds only for genes located on different chromosomes, or sufficiently far apart on the same chromosome. This chapter offers a comprehensive account of linkage, the tendency of genes on the same chromosome to be inherited together, and of recombination, the partial disruption of that association through crossing-over. The classic Lathyrus odoratus cross of Bateson, Saunders and Punnett is analysed with a worked chi-square test to demonstrate the first evidence of linkage, followed by Morgan's Drosophila melanogaster experiments, the concept of linkage groups, and their relationship to the chromosome theory of inheritance. The text explains complete and incomplete linkage, cis/trans configuration, the testcross, and the conversion of recombination frequency into map units (centiMorgans) with numerical examples. The cytological basis of crossing-over during meiosis (chiasmata, tetrads) is presented, the two-strand and four-strand models are compared and resolved through Neurospora tetrad analysis, and the distance-shortening effect of multiple and double crossovers is addressed alongside the Haldane mapping function. Three-point crosses are worked through step by step for gene order, region-specific recombination frequencies, the coefficient of coincidence and interference. The chapter closes with genetic versus physical maps, molecular markers, linkage disequilibrium mapping, GWAS and genomic selection.
