β-laktam Antibiyotikler
Özet
β-laktam grubu antibiyotikler, β-laktam halkası varlığı nedeni ile isimlendirilmiş bakterisidal etkili antibiyotik grubudur. Hem gram negatif hem de gram pozitif hücre duvarı sentezinden sorumlu penisilin bağlayan proteinlerin (PBP) transpeptidaz aktivitesini inhibe ederek peptidoglikan sentezini bozarlar. Bu antibiyotikler, β-laktam halkası birimine farklı aktif grupların girmesi ile antibiyotik spektrumlarındaki çeşitliliklerine göre sınıflandırılırlar. Bu sınıflandırmaya göre: Penisilinler, sefalosporinler, karbapenemler ve monobaktamlar grupları bulunmaktadır. Ek olarak β-laktam/β-laktamaz kombinasyonları etkinliklerini arttırmak amacı ile kullanıma alınmıştır.
β-lactam group antibiotics are a group of bactericidal antibiotics named for the presence of a β-lactam ring. They generally act by inhibiting the transpeptidase activity of penicillin-binding proteins (PBPs) which are responsible for both gram negative and gram positive cell wall synthesis therefore disrupting peptidoglycan synthesis. They are sub-classified according to the diversity of their antibacterial spectrums by the integration of different active groups into the β-lactam ring unit. This group includes penicillins, cephalosporins, carbapenems and monobactams. In addition, β-lactam/β-lactamase combinations have been developed to increase their effectiveness.
