Drug Metabolism in Geriatric Group
Özet
Aging is the process of time-related alteration and deterioration in physiological functions of higher multicellular organisms that occurs from adulthood to death. There are several hallmarks of aging, namely mitochondrial dysfunction, genomic instability, stem cell exhaustion, cellular senescence, telomere shortening, deregulated nutrient sensing, loss of proteostasis, epigenetic alterations. Aging is associated with disabilities and complications such as frailty, decline in muscle/fat ratio, loss of ability of fertilization, and the most common diseases, e.g., neurological, cardiovascular and metabolic diseases as well as cancers. Furthermore, as with neonates, the elderly become more susceptible to the impact of drugs through alteration in pharmacodynamics and pharmacokinetic events and blunted compensation mechanisms. The pharmacokinetic events involve of absorption, distribution, elimination and excretion of drugs used. In this review, the metabolism of drugs that are particularly used in common dermatological indications will be highlighted.
