Özet

Obezite epidemisinin artışında, geleneksel faktörlerin yanı sıra endokrin bozucu kimyasallar (EBK) arasında yer alan "obezojenler" adı verilen sentetik maddelerin rolü, Çevresel Obezojen Hipotezi kapsamında son yıllarda yoğun şekilde incelenmektedir. Özellikle prenatal ve neonatal gibi hassas gelişim dönemlerinde bu kimyasallara maruz kalmak, vücudun hormonal sinyalizasyon süreçlerini, iştah regülasyonunu ve temel metabolik hızını bozarak bireyleri yaşamın ilerleyen evrelerinde yağ kitlesi artışına ve obeziteye yatkın hale getirmektedir. Moleküler düzeyde PPARγ ve RXR gibi nükleer reseptörleri aktive eden obezojenik ajanlar, kök hücrelerin adipositlere (yağ hücrelerine) farklılaşmasını ve lipid birikimini doğrudan indüklemektedir. Günlük yaşamda yaygın olarak maruz kalınan başlıca obezojenler arasında plastik kaplarda, kozmetiklerde ve endüstriyel ürünlerde bulunan Tributiltin (TBT), Sentetik Östrojen (DES), Persistan Organoklorinler (POP), Bisfenol A (BPA), Ftalatlar, Polibromlu Difenil Esterler (PBDE) ve Parabenler yer almaktadır. Hayvan çalışmalarında bu maddelerin kalıcı kilo artışına yol açtığı kesin olarak kanıtlanmışken, insan odaklı epidemiyolojik araştırmalarda henüz çelişkili sonuçlar bulunmakla birlikte, obezite ve metabolik bozukluklarla aralarındaki pozitif korelasyon dikkat çekmektedir. Bu doğrultuda, küresel halk sağlığını korumak adına kanıtlanmış obezojenlerin üretimi, kullanımı ve maruziyet yollarına yönelik daha sıkı politikaların geliştirilmesi ve yasal düzenlemelerin hayata geçirilmesi büyük önem taşımaktadır.

The role of synthetic chemicals called "obesogens," which are a subset of endocrine-disrupting chemicals (EDCs), in the rising global obesity epidemic is being extensively investigated under the Environmental Obesogen Hypothesis. Exposure to these chemicals during critical developmental windows, particularly prenatal and neonatal periods, alters hormonal signaling, appetite regulation, and basal metabolic rate, thereby predisposing individuals to increased fat mass and weight gain in later life. At the molecular level, these obesogenic agents activate nuclear transcription regulators like PPARγ and RXR, directly inducing the differentiation of stem cells into adipocytes and promoting lipid accumulation. The most common obesogens encountered in daily life through consumer products, plastics, cosmetics, and industrial applications include Tributyltin (TBT), Diethylstilbestrol (DES), Persistent Organic Pollutants (POPs), Bisphenol A (BPA), Phthalates, Polybrominated Diphenyl Ethers (PBDEs), and Parabens. While animal experiments have conclusively demonstrated that developmental exposure to these substances leads to irreversible weight gain, human epidemiological data still present some conflicting results despite showing significant positive correlations with obesity and metabolic disorders. Consequently, establishing robust public health policies and legal regulations regarding the restriction and management of verified obesogens is essential to mitigate their widespread impact on global metabolic health.
